<jats:title>Abstract</jats:title><jats:p>A novel lumazine peptide, aspergilumamide A (<jats:bold>1</jats:bold>), as well as a known analog penilumamide (<jats:bold>2</jats:bold>), were isolated from the mycelia of a marine‐derived fungus <jats:italic>Aspergillus</jats:italic> sp. (33241), obtained from the mangrove <jats:italic>Bruguiera sexangula</jats:italic> var. <jats:italic>rhynchopetala</jats:italic> collected from the South China Sea. The structure of <jats:bold>1</jats:bold> was identified by comprehensive spectroscopic analysis, including 1D‐ and 2D‐NMR, ESI‐MS, and MS/MS experiments. The absolute configuration of <jats:bold>1</jats:bold> was determined by <jats:italic>Marfey</jats:italic>'s method.
