The preparation of superoxide dismutase (SOD), aldolase and proteinases by affinity chromatography from callus tissue Rauwolfa serpentina is described. SOD was isolated by metal chelate affinity chromatography, aldolase by using cellulose phosphate and proteinases by using haemoglobin-Sepharose 4B. The homogeneity of these preparations was analyzed by polyacrylamide gel electrophoresis (PAGE). Some properties were studied and molecular weights were estimated by sodium docylsulphate PAGE. The alkaloids of culture cells were separated by thin-layer chromatography. These cells lines of Rauwolfia serpentina were shown to give high yields of commercial quality ajmalin and are stable. © 1988.
