In continuation of our studies on the alkaloids of the bark of Alstonia venenata R. Br., we report the results of further investigation on this plant. From the fruits, two new indole alkaloids—echitovenidine and (+)-minovincinine—were isolated. Echitovenidine's structure (III) was determined via UV, IR, mass, and NMR spectra, reduction with zinc and methanolic sulfuric acid, hydrolysis (yielding β-dimethyl-acrylic acid), and comparison with echitovenine derivatives. (+)-Minovincinine, an amorphous solid with positive optical rotation, was identified as the optical antipode of minovincinine; its O-acetyl derivative matched echitovenine in UV, IR, mass spectra, TLC Rf values, and mixed melting point. The co-occurrence of alkaloids with identical skeletons but opposite rotational signs (echitovenine and (+)-minovincinine) in the same plant is noted.
