Lignans from the leaves of Premna recinosa (Verbenaceae)

Abstract

This study investigated the lignans from the leaves of Premna recinosa (Verbenaceae). Seven hundred and fifty grams of powdered leaves were extracted with cold ethanol for 2 weeks, and the resulting residue was fractionated using vacuum liquid chromatography (VLC) on silica gel with petroleum ether-EtOAc mixtures of increasing polarity. The 50-70% EtOAc fractions were combined and subjected to Sephadex LH-20 column chromatography to obtain a mixture of three lignans, which were further purified by repetitive preparative thin-layer chromatography (prep. TLC) using CHCl3-MeOH (9:1) to yield 2 mg of (+)-1-hydroxypinoresinol, 10 mg of (+)-lariciresinol, and 7 mg of (-)-seco-isolariciresinol. The structures of these compounds were established based on spectroscopic data. This is the first report of mono- and bisfuranoid lignans in the genus Premna; such compounds have previously been reported from other genera of the Verbenaceae. Nor-lignans of the type found in Premna latifolia were not detected.
