CHAETOGLOBOSINS A - F are cytotoxic [13]cytochalasans containing tryptophan units from Chaetomium globosum. The structures of chaetoglobosins A and B have been verified by X-ray analysis. This communication concerns the structures of four other metabolites, chaetoglobosins C, D, E, and F, isolated by chromatography of chloroform extracts of the mold cultured on polished rice grains. Chaetoglobosin D (III) has the same molecular formula (C32H36O5N2) as A and B, shows similar spectral properties, and its structure was proved by acetylation to B diacetate and formation from A with BF3-etherate. Chaetoglobosin C (IV), isomeric to A, B, D, was proposed based on spectral data, isomerization from A, and negative tetrazolium salt reaction. Chaetoglobosins E (V) and F (VI), dihydro derivatives of A - D, were analyzed via NMR, NOE, and spectral properties, with their structures (V, VI) proposed as the most preferable formulations. The structures III - VI are put forward based on chemical and spectral evidences and isomerization reactions, though further work is ongoing.
