Abstract

commercially available precoated plates of TLC and HPTLC quality (10 x 10cm, 10x 20cm, 20 x 20cm) with all types of sorbents may be used. The separation distance depends on the plate size. With special preparation of a 10 x 20 cm plate by scraping the layer in the form of a segment, the separation distance can be increased to 8.5 cm. The flow rate of the mobile phase can also be made linear in CLC, by scratching lines in the plate. SCLC can be used for the separation of complex biological matrices. The chromatogram can be observed during the separation with an integrated UY-lamp and, depending on the results, the mobile phase may be changed. In Table I, all analytical separation techniques realised with the ROTAC-HROM are compared, and the most important operating parameters are summarized. Since CLC and SCLC are forced flow techniques, the advantage of the small particle sizes of the HPTLC plates can be used over the whole separation distance. All described analytical methods using the MC and UMC allow a direct up-scaling for preparative separation and isolation (3). The new analytical methods realized with ROTACHROM can significantly improve efficiency of the analysis of naturally occurring compounds from plant extracts and preparations.
